The Lucky Star "No! Yes." meme originates from a scene in the show where the Miyuki, Kagami, and Tsukasa all try to open a door, but get shocked by static. The only who didn't get shocked opening the door was Konata. When she's about to open the door, Kagami, in english, says "No!" and Konata, also speaking in english, says "Yes." and opens the door.
The scene is very popular within Lucky Star fans, and in March 2008, the youtuber 5p00ki made a video entitled "YES YES YES YES YES". This is the first instance of the meme, and currently has over 245,000 views.
Many people either put a different "yes" over konata's, or put something behind the door. Here are a few examples:
